Amoxicillin is a semi-synthetic acid-resistant broad-spectrum penicillin antibiotic. Because of its broad-spectrum, acid-resistant and convenient preparation, its soluble powder, amoxicillin sodium injection, amoxicillin suspension for injection, amoxicillin oil for injection and amoxicillin long-acting injection oil have been widely used in veterinary clinic. With the continuous improvement of technology and the decrease of price, it has become one of the most widely used antibiotics in veterinary clinic in recent years. 1. Current situation of amoxicillin powder injection preparations:

Because there is no sterile amoxicillin raw material and the solubility of amoxicillin is low at present, amoxicillin sodium should be used for both human and animal amoxicillin powder needles, rather than amoxicillin. But because the raw material price of amoxicillin sodium is twice as high as that of amoxicillin, there are very few amoxicillin powder needles on the market at present. Most of them are replaced by amoxicillin + cosolvent or penicillin or Annai recently.

2. The difference of physicochemical properties between amoxicillin and amoxicillin sodium:

(1) The solubility in water is different from that in alcohol.

(2) The PH value of aqueous solution is different:

The third-party laboratory tests showed that the aqueous solution of amoxicillin was acidic and the aqueous solution of amoxicillin sodium was alkaline. Because of the low solubility of amoxicillin in water, some manufacturers add cosolvents to enhance its solubility, while the cosolvents mostly use sodium bicarbonate or sodium hydroxide, so the aqueous solution of amoxicillin + cosolvents and amoxicillin sodium is alkaline.

3. Identification of amoxicillin sodium powder

1) Take 0.5g amoxicillin sodium sample and dissolve it in 5ml water for injection. Use PH test paper to determine the PH value of the sample. If the PH value is below 7.0, the sample may be penicillin or analgin. If the PH value is between 8.0 and 10.0, the sample may be amoxicillin sodium or amoxicillin + cosolvent.

2) If the measured sample PH is between 8.0 and 10.0, add 0.5g sample into 60 ml pure alcohol, shake, observe for 15 minutes, see the solubility, if amoxicillin sodium can be dissolved in alcohol; otherwise, amoxicillin + cosolvent.

4. Advantages and disadvantages of amoxicillin sodium and amoxicillin + cosolvent powder injection:

1) Amoxicillin sodium is more soluble than amoxicillin + cosolvent.

2) Amoxicillin sodium has higher bioavailability and stronger pharmacodynamics than amoxicillin + cosolvent.

3) Fuller loading: Because of the addition of cosolvent in amoxicillin powder needle, the content of amoxicillin per unit weight decreases, that is, the content of amoxicillin in 0.5g amoxicillin + cosolvent may be only 0.3g or 0.4g.
